Nanoemulsions
A major challenge in the race to bring innovative treatments, therapies and cures to market is that many of the drugs now being developed, including those using cannabis oil, are highly water-insoluble and exhibit poor bioavailability. A solution is to create nanoemulsions, also known as submicron emulsions or mini-emulsions. These non-toxic, liquid oil-in-water or water-in-oil formulations produce tiny droplets (50-1000 nm) that are generally recognized by the FDA as safe for human consumption, skin penetration, eye application, etc.

But to create viable products, you need scalable processing equipment that consistently and uniformly produces nano-sized particles and distributes them evenly in the nanoemulsion. Dyhydromatics gives you that competitive advantage. Our laboratory processors are directly scalable to our pilot/production scale processors due to its unique reaction chamber technology.

Dyhydromatics ShearJet processors make it easy to fabricate highly stable nanoemulsions batch after batch. As the initial macroemulsion flows through the ShearJet Diamond Reaction Chamber, each mL experiences the same high shear rates. This produces uniform and precisely-sized nano-droplets without using solvents.. Fewer passes reduce your processing time and costs. Plus, the uniformity of the nano-particles allows you to sterile-filter your nanoemulsions which can significantly lower your sterilizing expenses.

The ShearJet processors are designed to overcome several other nanoemulsion production challenges. Each comes with a simple-feed, jacketed, helicoil heat exchanger which reduces temperatures that may trigger denaturing or damage to the microemulsions. You will never have to refill equipment ice compartments during the process again. The ShearJet’s pharma-grade parts are easy to clean between batches which helps you meet the most stringent cGMP sanitation protocols. Plus, the portability of the HL60 processor makes it an economical solution when producing multiple medicines in multiple rooms daily. Simply move the processor from room to room.

Cell Disruption
If you are charged with overseeing cell disruption at your facility, one of your biggest challenges is acquiring equipment that gently ruptures the highest number of cells so you can harvest the greatest amount of soluble protein, nucleic acids or natural products . Moreover, many soil-dwelling microorganisms have the ability to produce secondary metabolites, also referred to as natural products, that give them the advantage over other microorganisms in a highly competitive environment, such as soil. They exhibit various biological activities such as anticancer, antifungal, antibacterial, immunosuppressive, protein inhibitory, etc. If those biosynthesized natural products are retained in the cell, the cells have to be disrupted in order to access the valuable molecules. Depending on the chemical properties of the molecules and consequently the downstream processing steps, high pressure homogenizator can be a valuable addition to such process. Typically, as you increase the rupture rates you also increase processing temperatures and/or pressure levels during cell lysing. These higher temperatures and pressure levels can damage or denature the sought-after cellular contents costing you valuable time and wasted product.
Now there is a scalable solution for your pharmaceutical cell lysing applications. Our laboratory processors are directly scalable to our pilot/production scale processors due to its unique reaction chamber technology.

With Dyhydromatics ShearJet processors you can consistently achieve cell disruption efficiency of 99%-plus and harvest more valuable cell material from mammalian, yeast, bacterial (E. coli) and algae cells. As the cells flow through the ShearJet Diamond Reaction Chamber, each experiences the same high shear rates. The resulting large cell membrane fragments make it easier to separate the cell products from the debris downstream than with a French press, mortar and pestle or other brands of high-pressure homogenizers. The ShearJet processing seldom changes the viscosity of the solution, a problem that also occurs when using other cell lysing equipment or techniques.

Each Dyhydromatics processor comes with a simple feed, jacketed , helicoil heat exchanger that effectively cools the product in the Diamond Reaction Chamber so there is little to no damage or denaturing of the molecules. You will never have to use ice to chill your lysing process again. This, plus fewer passes through the chamber, gives you tighter control over the pressure levels, temperature and shearing forces that impact the quality of your harvested cell products.

The ShearJet’s pharma-grade parts are easy to clean between runs which helps you meet the most stringent cGMP sanitation protocols. Also, you will find the portable HL60 processor is an economical solution when your daily schedule demands lysing multiple types of cells in multiple rooms. Just put the processor on a cart and move it throughout your facility.

Liposomes
Using liposomes for new drug delivery systems is an exciting area of research and manufacturing in the pharmaceutical industry. These artificially-created microscopic and spherical cells composed of the main cell membrane components, phospholipids, can solubilize drugs with low aqueous solubility, protect them from the breakdown and so prolong their residence time in the body. Those lipid nanoparticles due to their small size and body-friendly composition evade the body’s biological gatekeepers and reach their target cell without being degraded. Additionally, liposomes give drugs a greater therapeutic index and improve their bioavailability due to controlled release in the targeted tissues. But whether you are producing liposomes to stealthily deliver chemotherapeutic drugs, antifungal agents or mRNA molecules for anticancer treatments or coronavirus vaccine, one of your challenges is to minimize the volume and cost of solvents as you scale up production.
Dyhydromatics ShearJet processors generate high-shear forces and this is typically sufficient for production of high-quality liposomes. That means you can spend less on solvents (and less time removing them) when you scale up your production with these high shear processors. Our laboratory processors are directly scalable to our pilot/production scale processors due to its unique reaction chamber technology.

At all production levels, the ShearJet’s Diamond Reaction Chamber applies the same high-shear rate to every mL. This allows you to consistently produce high quality, uniform, nano-sized liposomes in the quantities you want. Each ShearJet model comes with a simple feed, jacketed , helicoil heat exchanger that effectively cools your product in the Diamond Reaction Chamber so there is little-to-no denaturing or damage to your liposomes and therapeutic agents. This, plus fewer passes through the chamber, gives you tighter control over the pressure levels, temperature and shearing forces batch after batch.
The ShearJet’s pharma-grade parts are easy to clean between runs so you can meet some of the most stringent cGMP sanitation protocols. Also, the portability of the HL60 processor makes it an economical addition to your facilities. You simply move it from room to room as you make multiple types of liposomes throughout the day.

Polysaccharide Nanoparticles
Pathogenic bacteria commonly produce a thick, mucous-like, layer of polysaccharide that as a capsule covers surface carbohydrates and proteins that would otherwise provoke an immune response and lead to the destruction of the bacteria. Those capsular polysaccharides are incredibly structurally diverse, even among one bacterial species such as E. coli, and mixtures of polysaccharides, either conjugated or native are used as vaccines and adjuvants. Globally used is a pneumococcal polysaccharide vaccine (PPV) that protects against a few different bacterial variations. As vaccines are injected into the human body, they undergo a sterile filtrations step, which requires the particles to have less than 200 nm. Polysaccharides are due to their biocompatibility and biodegradation properties also used as drug delivery systems.

Nanoencapsulations
In addition to biological encapsulations such as nanoemulsions, liposomes and polysaccharides, other materials are being tested for drug delivery in various applications. Many drugs have been encapsulated on polymeric micelles, which due to their small size and high structural stability, enhance their solubility and therapeutic activity. Micelles can be formed by hydrophobic interactions, polyion complex micelles by electrostatic interactions and micelles by metal ion complexation. Polymeric micelles were shown to enhance the biological activities of the conjugated drugs such as greater cytotoxicity to cancer cells, due to accurate targeting, and show reduced systemic uptake of the drug to normal cells, slow and sustained release, which is in some cases even pH-dependent. For example, various materials are being tested for efficiency in oral administration of insulin. A peptide hormone present in blood is instantly affected by acidic environment in the stomach and digestive enzymes present throughout the gastrointestinal tract (GIT). However, lipid nanoparticles e.g., niosomes, archeosomes, and ionic nanoparticles, e.g., gold, iron oxide, calcium phosphate, zirconium phosphate (ZrP) nanoparticles, with encapsulated insulin enhance drug absorption in the GIT, increase GIT residence time, protect the loaded insulin from chemical and enzymatic degradation, and gradually release insulin from the lipid matrix into blood, resulting in improved therapeutic action.

Suspensions and Dispersions Pharmaceutical
The active pharmaceutical ingredients produced by bacteria or plants are usually extracted, purified and dried, resulting in solid particles. Nanosuspensions and nano-dispersions, consist of finely dispersed solid drug particles in a dispersant - aqueous or organic vehicle, stabilized with a stabilizer, that have their size greatly reduced. The nano-size increases:
-
the drug´s solubility,
-
bioavailability and
-
lowers the adverse effects, due to its rapid dissolution and tissue targeting.
Such nanosuspensions, consisting of water-soluble or poorly water-soluble solid particles, can also be incorporated into the tablets. The examples of such nanosuspensions are the antibiotic azithromycin from Sandoz, the antifungal medication amphotericin, the cholesterol lowering drug fenofibrate nanosuspension and a promising anticancer agent curcumin.
If you are dealing with suspensions yourself and you are interested in scalable processing equipment that consistently and uniformly produces nano-sized particles, you came to the right place. Dyhydromatics gives you that competitive advantage. Dyhydromatics Reaction Chambers are made of diamond, which extends their lifespan compared to chambers made of ceramics. They are available in different sizes and can be multi-slotted, which ensures that the process developed with a single-slotted Reaction Chamber is scalable.
Dyhydromatics ShearJet processors make it easy to reduce the particles size of the processed formulation, fabricating nano-sized particles batch after batch. As the initial pre-processed formulation flows through the ShearJet Diamond Reaction Chamber, each mL experiences the same high shear rates. This produces uniform, precisely-sized nano-particles that can be achieved in fewer passes, which reduces your processing time costs.
